        <title>test from IPT Salza dataset - Version 9</title>

        <description>This dataset containts vegetation plots that were collected specifically for the purpose of the PhD thesis of Salza Palpurina defended at Masaryk University in 2017 (Palpurina, 2017). Together with colleagues from the Vegetation Science Group at Masaryk University and from Sofia University, we sampled an initial dataset of 181 vegetation plots in dry grasslands across Bulgaria and Dobrudzha in the years 2010–2012. Visiting time was during the peak of growing season for dry grasslands in this area, i.e. end of May till beginning of July. We used a protocol for soil and vegetation sampling that matched the protocol employed for field expeditions by Vegetation Science Group at Masaryk University (see the section on Sampling methods). A map and a detailed description of the study area are provided in Palpurina et al. (2015). The dataset with full species composition is stored in the Balkan Dry Grasslands Database, which is registered in the Global Index of Vegetation-Plot Databases (http://www.givd.info; Dengler et al., 2011) under the code EU-00-013. However, the current dataset is the most up to date in terms of species composition as it also contains updates as a result of revised specimens linked to occurrences from respective vegetation plots.</description>

        <description>Meise Botanic Garden (MeiseBG) has a history that goes back to 1796. Today, it is an internationally recognized botanic garden in a park of 92 hectares, and a centre of excellence for plant biodiversity research. MeiseBG houses the 15th largest herbarium in the world, holding 4 million preserved specimens, a comprehensive botanical library, a seed bank and a living plant collection with 25,000 accessions of plants from around the world. Research focuses on plant, algal and fungal taxonomy, evolution, biodiversity conservation, ecosystems and ethnobotany. The preserved collections (including the herbarium, wood, carpological, slide and molecular collections) have a global scope, with a focus on Central Africa, Belgium, and Southwestern Europe, with additionally important historic collections from Latin America. Highlights are the private collections of famous 19th botanists such as Henri Van Heurck (diatoms), Carl Von Martius (Flora brasiliensis), Heinrich Gustav von Reichenbach (orchids) and Crépin (wild roses), which form the historic core of the collections. A wide range of taxonomic groups are covered including: vascular plants, lichens, mosses, liverworts, fungi, myxomycetes, macroalgae, and diatoms. Meise Botanic Garden is dedicated to digitally unlock these precious and unique botanical collections.</description>

        <title>Insektmobilen - National citizen science and DNA metabarcoding survey of flying insects in June 2018 and 2019 (TEST REGISTRATION) - Version 3</title>

        <description>The Insectmobile (Insektmobilen) is a research project at the National History Museum of Denmark, University of Copenhagen, with the goal to investigate the diversity of flying insects in Denmark. In the summer of 2018, 2019 and 2020 almost 400 volunteers collected flying insects using large custom made insect nets mounted on the roof of their cars. The bulk insect samples were processed with a non-destructive DNA extraction DNA metabarcoding protocol (dx.doi.org/10.17504/protocols.io.bmunk6ve) and sequences were assigned taxonomy by importing the fasta file into GBIF&#39;s sequence ID tool (https://www.gbif.org/tools/sequence-id). The sequences were queried against a 99% clustered version of the BOLD Public Database v2024-01-06 public data (COI-5P sequences).

The dataset contains unidentified sequences and potential errors and contaminants. For example, even though the primers used were developed as universal primers targeting freshwater insects, you will find other phyla, classes etc. We share these sequences and associated data for the data to be as open as possible, but please do reannotate sequences and filter the data for your specific needs prior to using the data for analysis. Please be aware that the samples may contain gut content of sampled insects and eDNA.

Sequence identification certainty is captured in the identificationRemarks field. The bit score is the required size of a sequence database in which the current match could be found just by chance. The bit score is a log2 scaled and normalized raw score. Each increase by one doubles the required database size (2bit-score). The expect value is a parameter that describes the number of hits one can &quot;expect&quot; to see by chance when searching a database of a particular size. It decreases exponentially as the score of the match increases. Hence, a low expect value is better. How much of the query(input) sequence aligns with the match in the the reference database, in percent. Badges representing different identity thresholds (match types); Blast exact match = identity &gt;= 99% and queryCoverage &gt;= 80%. This is within the threshold of the OTU, Blast ambiguous match = identity &gt;= 99% and queryCoverage &gt;= 80%, but there is at least one more match with similar identity, Blast close match = identity &lt; 99% but &gt; 90% and queryCoverage &gt;= 80%. It is something close to the OTU, maybe the same genus, Blast weak match = there is a match, but with identity &lt; 90% or/and queryCoverage &lt; 80%. Depending on the quality of the sequence, bit score, identity and expect value, a higher taxon could be inferred from this, Blast no match = no match to the reference database.         <title>Integrated Dataset of Fungi Associated with Peltigera Cyanolichens from Southern Chile - Version 2</title>

        <description>This integrated dataset compiles fungal diversity data associated with Peltigera cyanolichens collected across southern Chile by the Microbial Ecology Lab at Universidad de Chile. It merges three occurrence resources: (1) a collection of over 1,000 Peltigera lichen thalli sampled from diverse environments, with molecular identification of the main symbionts; (2) 180 basidiomycete yeasts; and (3) 45 ascomycete fungi, both isolated from a subset of the previously collected Peltigera thalli. The isolates were identified by sequencing the ITS marker and through subsequent phylogenetic analyses.

The purpose of creating this integrated resource was to test and explore the structure and flexibility of the new Darwin Core data format, using a case study that links macro- and micro-organismal diversity within lichen symbioses.
